Electrochemical detection of redox molecules secreted by Pseudomonas aeruginosa – Part 1: Electrochemical signatures of different strains

چکیده
During infections, fast identification of the microorganisms is critical to improve patient treatment and better manage antibiotics use. Electrochemistry exhibits several advantages for rapid diagnostic: it enables easy, cheap in situ analysis redox molecules most liquids. In this work, culture supernatants different Pseudomonas aeruginosa strains (including PAO1 its isogenic mutants PAO1ΔpqsA, PA14, PAK CHA) were analyzed by square wave voltammetry on glassy carbon electrode during bacterial growth. The obtained voltamograms shown complex traces exhibiting numerous peaks with potential repartitions current amplitudes depending studied bacterium and/or growth time. Among them, some clearly associated well-known toxin Pyocyanin (PYO) autoinducer Quinolone Signal (PQS). Other observed that are not yet attributed known secreted species. Each electrochemical response (number peaks, peak amplitude) can be interpreted as a fingerprint or “ID-card” strain may implemented bacteria identification.

The Direct Electrochemical Investigation of Redox Enzymes
Even though the use of enzymes as useful analytical reagents when immobilized onto a transducer such as an electrochemical device has been reported in the literature for nearly two decades, the direct, i.e. unmediated electrochemistry of enzymes is, nevertheless, a rather recent development. Efforts will be made to introduce the fundamental basis for protein-surface interactions with emphas...
متن کاملElectrochemical detection of single molecules.
The electrochemical behavior of a single molecule can be observed by trapping a small volume of a dilute solution of the electroactive species between an ultramicroelectrode tip with a diameter of approximately 15 nanometers and a conductive substrate. A scanning electrochemical microscope was used to adjust the tip-substrate distance ( approximately 10 nanometers), and the oxidation of [(trime...

متن کاملMonitoring of Pseudomonas aeruginosa Toxins via Miniaturized Electrochemical Assemblies
The production of pyocyanin from the opportunistic pathogen Pseudomonas aeruginosa was probed using a variety of miniaturized electrochemical systems. Goal 1 used disposable screen printed carbon electrodes to measure pyocyanin in medically relevant samples showing for the first time that pyocyanin can be detected at medically relevant concentrations (1-100 µM) without sample processing. Goal 2...
متن کاملAntimicrobial Resistance Profile of Pseudomonas Aeruginosa Strains
Abstract Background and Objectives: Pseudomonas aeruginosa is the most common organism, which is separated from the burn infections. Due to increased antibiotic resistance, there are many problems to deal with the infections caused by Pseudomonas aeruginosa. This study aimed to determine the resistance to antibiotics against clinical isolates of Pseudomonas using phenotype me...
